| 有效的激發,
分光, 及 光譜鑑別蒐集, 是必須依賴您的樣品及螢光染劑而決定之.
如上圖, 左邊圖示, 因為 Emission
filter 的設計欠佳, 以致產生螢光重疊雜訊.
右圖圖示, 表示適當正確的 Emission
filter 規劃, 產生紅綠光譜分離的影像. |

活體螢光蛋白 (
Living color protein ) 的光譜都極為接近, 在使用上,
要謹慎處理選擇濾鏡波長的規劃. 取自 Clontech. |

| 螢光濾鏡

|
|
|
|
每一組螢光濾鏡基本上都包括了三個鏡片
1).
激發光濾鏡
(
Excitation Filter )
因為特定的染劑需要使用特定波長範圍的光來當作激發光源
( 才不致影響到其它不想要檢視的物質被激發
) 而一般燈泡式的光源
( 如
Hg
50W, Hg 100W, Xe 75 W ), 所產生的光,
都含蓋全波長
(
300 – 1200 nm ) , 所以,
必須使用激發光濾鏡來篩選激發螢光染劑所須的光譜範圍
(
可為
Long-pass,
Band-pass, Short-pass ). 激發光濾鏡是以特殊的鍍膜,
來產生可篩選波長的功能.
例如
BP
450 – 490 nm, 即是僅讓
450
– 490 nm 間的光通過,
用以激發
FITC
染劑.
2).
分光鏡
(
Dichromatic Mirror 或者
Beam
Splitter )
將較短波長的激發光反射至樣品.
例如
RKP
510 nm, 即是,
510 nm 以下的光反射至樣品,
然而,
510 nm 以上的光可通過.
3).
釋放光濾鏡
(
Emission Filter 或者
Suppression
Filter, Barrier filter )
受檢樣品中的特定染劑及其它物質被激發後,
所有被激發的物質都釋放出來較長波長的螢光,
然而螢光僅能在釋放光濾鏡所選定的特定波長範圍內通過,
此通過的螢光即是我們所要觀察的螢光影像.
例如
:
LP 510 nm,
即是,
螢光光譜超過
510
nm 的,
都能通過濾鏡供觀察,
低於
510
nm 的螢光則被阻擋看不到.
