Centromeric probes also called: CEP probes (Chromosome Enumeration
Probes)

 

The first probes used for FISH analysis comprised of DNA sequences for
the centromeric area of each chromosome. This region consists of a sequence
of DNA that is repeated hundreds of times. This class of DNA probes is
suitable for the analysis of numerical aberrations such as trisomy (the
presence of an extra chromosome) and monosomy (the absence of a single
chromosome). 

One of the important advantages of FISH over conventional cytogenetic
techniques is the ability to visualize the signals through all stages of
the cell cycle and not only in dividing cells at the metaphase stage. This
approach is termed "interphase cytogenetics". 

The advantages of "interphase FISH" for the diagnosis of numerical
aberrations are obvious. The technique is fast and the signals can be
visualized on different kinds of cells : fetal cells, embryonic cells,
cancer cells and more. The signals are easily detected compared to the
assessment of chromosomal bands. However, since the whole karyotype is
not visible, we can detect only specific aberrations according to the
probes that were used in the analysis. In tumor analysis interphase
aberrations detection is of great importance due to the difficulty in
producing metaphase spreads that are representative of the whole
population. Therefore, FISH has already become an important tool in tumor
analysis for the detection of numerical aberrations that are of prognostic
important. 
unique DNA sequences also called LSI (Locus Specific Identifier Probes)
The second group of probes are the unique sequences probes. These are
single copy sequences that can range from very small probes (approx. 1 Kb-
1000 base pairs of DNA) to a very large ones which consist of hundreds of
Kb. These region specific probes can be used for diagnosis of specific
translocation, cryptic translocation in the telomeres and detection of
deletions in prenatal diagnosis and in cancer analysis. These probes can be
used either in interpahse or in metaphase.
painting probes also called WCP (Whole Chromosome Paints Probe)
The third group of probes are the painting probes. These are composite
probe sets specific for the entire chromosomes. This type of probes enable
the whole chromosome to be stained. The painting probes are mainly useful
for the detection of structural chromosomal aberrations like translocation
only in metaphases. Painting probes have also proven useful for the
identification of marker chromosomes (that is a chromosome that is
different from any normal chromosome in the karyotype). 

The use of painting probes is also very useful for the detection of
reciprocal translocations after exposure to ionizing radiation., in
mutagenesis studies and chemical dosimetry.

ASI can supply right away all the 24 individual human chromosomes or
the 21 mouse chromosomes labeled in Green or in Red. We can supply each one
in a separate kit for 10 slides or we can supply them in combination of two
chromosomes together: one in Red and one in Green. We can also supply a kit
with three chromosomes: one in Red , one in Green and one in Yellow.
