LEICA DMi8 S Module Infinity TIRF 全反射螢光顯微影像系統 (TIRFM), 架構在 DMi8 S 倒立顯微鏡上, 隸屬物鏡式全反射螢光顯微鏡, 全自動調整激發光的入射角度大於臨界角, 可達到奈米等級解析度和高訊號/背景值的特性,廣泛應用於細胞表面螢光標定的極薄區域物質的動態觀察 (動態範圍通常在200nm以下). 可觀察溶液態中的單分子螢光影像和光譜, 多色螢光影像, 超解析奈米影像. 單分子動態追蹤,  GSD, dSTORM, uPaint., 量測螢光共振能量轉移 (FRET)、螢光半衰期成像 (FLIM).

獨家超廣視野的 PL-APO 100x/1.46 TIRF 物鏡, 視野數達 (FOV) 到 19 mm. 可探索臨界膜的超細為分子結構與動態反映.

LEICA Infinity TIRF 可在 405 - 640 nm 波長範圍內, 可選用 1 - 5 支固態雷射光源. AOTF 調控雷射強度.

LEICA Infinity TIRF 架構在 DMi8 S 倒立顯微鏡, 採用獨特的主動式自動對焦系統, 軟體採用 LEICA LAS X.

 





Leica Infinity TIRF  光路設計 與 TIRF 技術原理

TIRF (Total Internal Reflection Fluorescence) 利用雷射光入射角度達到全反射時,在介質表面形成的漸逝波(Evanescent Wave)做為激發螢光的能量來源,由於漸逝波的深度僅於表面 70~300 nm,較深的螢光物質便不會被激發,只有極靠近全反射面的樣本區域會產生螢光反射,大大降低了背景光雜訊干擾觀測標的,故此項技術廣泛應用於細胞表面物質的動態觀察。因此可觀察表面螢光表現而不受干擾 ( 例如細胞膜內外的動態觀察 ),為提高 Z 軸解析度之一項技術.

全內反射螢光顯微術 ( TIRFM) 是探索細胞基底的淺層接觸區域內的細胞生命活動現象的有效光學探測方法.,如細胞膜內蛋白質的動力學過程,基底附近的細胞骨架,細胞運動…等, 可應用於即時觀察單一肌漿球蛋白分子的運動、單一蛋白分子對之間的螢光共振能量轉移 (FRET),…..酶的轉譯,聚合物內單個分子的結構變化, 等離子體膜附近的神經分泌腺的顆粒運動.
